利用超臨界流體的溶解能力與其密度的關(guān)系,即利用壓力和溫度對(duì)超臨界流體溶解能力的影響而進(jìn)行的。在超臨界狀態(tài)下,將超臨界流體與待分離的物質(zhì)接觸,使其有選擇性地把極性大小、沸點(diǎn)高低和分子量大小的成分依次萃取出來(lái)。當(dāng)然,對(duì)應(yīng)各壓力范圍所得到的萃取物不可能是單一的,但可以控制條件得到最佳比例的混合成分,然后借助減壓、升溫的方法使超臨界流體變成普通氣體,被萃取物質(zhì)則完全或基本析出,從而達(dá)到分離提純的目的,所以超臨界CO2流體萃取過(guò)程是由萃取和分離過(guò)程組合而成的。
超臨界CO2是指處于臨界溫度與臨界壓力(稱(chēng)為臨界點(diǎn))以上狀態(tài)的一種可壓縮的高密度流體,是通常所說(shuō)的氣、液、固三態(tài)以外的第四態(tài),其分子間力很小,類(lèi)似于氣體,而密度卻很大,接近于液體,因此具有介于氣體和液體之間的氣液兩重性質(zhì),同時(shí)具有液體較高的溶解性和氣體較高的流動(dòng)性,比普通液體溶劑傳質(zhì)速率高,并且擴(kuò)散系數(shù)介于液體和氣體之間,具有較好的滲透性,而且沒(méi)有相際效應(yīng),因此有助于提高萃取效率,并可大幅度節(jié)能。
超臨界CO2的物理化學(xué)性質(zhì)與在非臨界狀態(tài)的液體和氣體有很大的不同。由于密度是溶解能力、粘度是流體阻力、擴(kuò)散系數(shù)是傳質(zhì)速率高低的主要參數(shù),因此超臨界CO2的特殊性質(zhì)決定了超臨界CO2萃取技術(shù)具有一系列的重要特點(diǎn)。超臨界CO2的粘度是液體的百分之一,自擴(kuò)散系數(shù)是液體的100倍,因而具有良好的傳質(zhì)特性,可大大縮短相平衡所需時(shí)間,是高效傳質(zhì)的理想介質(zhì);具有比液體快得多的溶解溶質(zhì)的速率,有比氣體大得多的對(duì)固體物質(zhì)的溶解和攜帶能力;具有不同尋常的巨大壓縮性,在臨界點(diǎn)附件,壓力和溫度的微小變化會(huì)引起CO2的密度發(fā)生很大的變化,所以可通過(guò)簡(jiǎn)單的變化體系的溫度或壓力來(lái)調(diào)節(jié)CO2的溶解能力,提高萃取的選擇性;通過(guò)降低體系的壓力來(lái)分離CO2和所溶解的產(chǎn)品,省去消除溶劑的工序。
在傳統(tǒng)的分離方法中,溶劑萃取是利用溶劑和各溶質(zhì)間的親和性(表現(xiàn)在溶解度)的差異來(lái)實(shí)現(xiàn)分離的;蒸餾是利用溶液中各組分的揮發(fā)度(蒸汽壓)的不同來(lái)實(shí)現(xiàn)分離的。而超臨界CO2萃取則是通過(guò)調(diào)節(jié)CO2的壓力和溫度來(lái)控制溶解度和蒸汽壓這2個(gè)參數(shù)進(jìn)行分離的,故超臨界CO2萃取綜合了溶劑萃取和蒸餾的2種功能和特點(diǎn),進(jìn)而決定了超臨界CO2萃取具有傳統(tǒng)普通流體萃取方法所不具有的優(yōu)勢(shì):通過(guò)調(diào)節(jié)壓力和溫度而方便地改變?nèi)軇┑男再|(zhì),控制其選擇性;適當(dāng)?shù)剡x擇提取條件和溶劑,能在接近常溫下操作,對(duì)熱敏性物質(zhì)可適用;因粘度小、擴(kuò)散系數(shù)大,提取速度較快;溶質(zhì)和溶劑的分離徹底而且容易。
超臨界流體萃取技術(shù)是目前世界上公認(rèn)的一種高科技生物分離技術(shù)。具有低溫、無(wú)毒、分離效率高等特點(diǎn)。萃取過(guò)程由萃取和分離組合而成。
